HYL1 is a nuclear protein involved in the processing of miRNAs but its exact function remains unknown. Arabidopsis thaliana hyl1 mutants exhibit hypersensitivity to ABA. We decided to answer the question whether ABA affects the HYL1 protein localization within the cell and show that it does not. We also studied the expression of HYL1 in different tissues and organs. In this paper we show for the first time the expression profile of the HYL1 protein using anti-HYL1 antibodies. The protein is present in seedlings and mature plants in all organs studied, with the highest amount in inflorescences. A. thaliana HYL1 protein has several repetitions of a 28-amino-acid sequence at the C-terminus that confer protein instability. Our bioinformatic analysis of HYL1 homologs in different Brassica species shows that this repetition is typical only for Arabidopsis. This may suggest a relatively late evolutionary acquisition of the C-terminal domain.  相似文献

Field research on grassland fungi in Slovakia resulted in collection of several rare fungi, including also geoglossaceous taxa, such as Microglossum viride (5 sites), Trichoglossum hirsutum (13 sites), Trichoglossum walteri (5 sites), Trichoglossum variabile (1 site), Microglossum olivaceum (2 sites), Thuemenidium atropurpureum (1 site). Tissue culture systems have long been exploited to study the process of organogenesis. In response to externally applied cytokinins, pluripotent cells proliferate into green calli and subsequently regenerate shoots. Conventionally, the cytokinin-induced greening phenotype has been evaluated by counting numbers of green foci or to present photographic evidence of morphological changes. However, because the structure of calli is disorganized and the development of pigmentation takes place gradually from pale white through yellow to green, adequately defining and counting green foci remains difficult. In this study, we employed chlorophyll measurement as an alternative method to statistically assess the greening phenotype in tissue culture material. We found that N,N-dimethylformamide was the most effective solvent for the extraction of chlorophylls from callus tissue and that bead disruption of the structured tissue improved solvent penetration and the consistency of results. The sensitivity of the method facilitated the quantification of chlorophylls in single-cultured root explants and the use of a spectrophotometer increased the efficiency of measuring multiple samples. Our measurements showed that chlorophyll contents from calli of wild-type and altered cytokinin response mutants (cre1; ahk3, or cytokinin hypersensitive 2 (ckh2)/pickle (pkl) were statistically distinguishable, validating the method. Our proposed procedure represents gains in efficiency and precision and leads to more robust standardization than the conventionally used counting of green foci.

Testing German shepherd puppies to assess their chances of certification   总被引:1,自引:0,他引:1  
Behavioral activity of 7-week-old German shepherd puppies was tested and the activities analyzed if they could be used for predicting police efficiency of the individual. In total 206 individuals sired by 42 sires and 44 dams were used. The activities were divided into 10 tasks in which reactions and behavior of pups were scored from 0 to 5 points. All pups were tested separately from other conspecifics. Probability that the puppy will pass the certification was tested by a logistic regression. Of the 206 puppies, 148 passed the certification while 58 failed. Some tested behavioral variables were moderately to highly correlated with one another. Therefore we applied a factor analysis. Three factors were retained accounting for 100% of the shared variance. After inspection of the rotated factor pattern matrix and its confidence intervals, it appeared that variables “Independent movement and interactions with the tester”, “Negotiating obstacles”, “Entering a room”, “Behavior toward a person”, and “Behavior in new environments” loaded on Factor 1 (“Factor for movement”), while variables “Response to distracting stimuli caused by a shovel”, “Response to a distracting noise while left alone in a room”, and “Response to loud distracting stimuli” on Factor 2 (“Factor for responding to noise”) and variables “Retrieval” and “Tug of war” on Factor 3 (“Factor for attitude to predation”). In the final logistic regression model, the probability that the puppy will pass the certification depended on the higher weight at the time of testing (, P = 0.0005), on the “Factor for attitude to predation” (, P = 0.0007), on the “Factor for responding to noise”, where the higher the score, the weaker was the response (, P = 0.0232), and on the “Factor for movement” showing an increasing probability with decreasing score (, P = 0.0219). The tests in our study seem to be a good base which might enable selection for suitable dogs as early as 7 weeks of age. The puppies having high probability to pass certification in adulthood were heavy individuals willing to chase, catch, and fetch a tennis ball, and follow a rag drawn away from them, while weakly responding to a distracting noise in various situations and showing low activity while negotiating obstacles and moving and interacting with the tester. To conclude, contrary to skeptical assumptions expressed by various authors, the specific puppy tests for police dogs provide a reliable tool for predicting future service ability of a puppy. Differences in methodology are likely to contribute to a lack of consensus among various studies.  相似文献

Simian immunodeficiency virus (SIV) persistence in wild populations of African nonhuman primates (NHPs) may occur through horizontal and vertical transmission. However, the mechanism(s) and timing of the latter type of transmission have not been investigated to date. Here we present the first study of SIV transmissibility by breast-feeding in an African NHP host. Six mandrill dames were infected with plasma containing 300 50% tissue culture infective doses of SIVmnd-1 on the day after delivery. All female mandrills became infected, as demonstrated by both plasma viral loads (VLs) and anti-SIVmnd-1 seroconversion. Neither fever nor lymphadenopathy was observed. At the peak of SIVmnd-1 viral replication (days 7 to 10 postinoculation), plasma VLs were high (8 x 10(6) to 8 x 10(8) RNA copies/ml) and paralleled the high VLs in milk (4.7 x 10(4) to 5.6 x 10(5) RNA/ml). However, at the end of the breast-feeding period, after 6 months of follow-up, no sign of infection was observed for the offspring. Later on, during a 4-year follow-up examination, two of the offspring showed virological evidence of SIVmnd-1 infection. Both animals seroconverted at least 6 months after the interruption of lactation. In conclusion, despite extensive viral replication in mandrill mothers and high levels of free virus in milk, no SIVmnd-1 transmission was detectable at the time of breast-feeding or during the following months. Since we observed a markedly lower expression of CCR5 on the CD4(+) T cells of young mandrills and African green monkeys than on those of adults, we propose that low levels of this viral coreceptor on CD4(+) T cells may be involved in the lack of breast-feeding transmission in natural hosts of SIVs.  相似文献

The mammalian epigenetic phenomena of X inactivation and genomic imprinting are incompletely understood. X inactivation equalizes X-linked expression between males and females by silencing genes on one X chromosome during female embryogenesis. Genomic imprinting functionally distinguishes the parental genomes, resulting in parent-specific monoallelic expression of particular genes. N-ethyl-N-nitrosourea (ENU) mutagenesis was used in the mouse to screen for mutations in novel factors involved in X inactivation. Previously, we reported mutant pedigrees identified through this screen that segregate aberrant X-inactivation phenotypes and we mapped the mutation in one pedigree to chromosome 15. We now have mapped two additional mutations to the distal chromosome 5 and the proximal chromosome 10 in a second pedigree and show that each of the mutations is sufficient to induce the mutant phenotype. We further show that the roles of these factors are specific to embryonic X inactivation as neither genomic imprinting of multiple genes nor imprinted X inactivation is perturbed. Finally, we used mice bearing selected X-linked alleles that regulate X chromosome choice to demonstrate that the phenotypes of all three mutations are consistent with models in which the mutations have affected molecules involved specifically in the choice or the initiation of X inactivation.  相似文献

Children with human immunodeficiency virus infection often have higher viral loads and progress to AIDS more rapidly than adults. Since the intestinal tract is a major site of early viral replication and CD4(+) T-cell depletion in adults, we examined the effects of simian immunodeficiency virus (SIV) on both peripheral and intestinal lymphocytes from 13 neonatal macaques infected with SIVmac239. Normal neonates had more CD4(+) T cells and fewer CD8(+) T cells in all tissues than adults. Surprisingly, neonates had substantial percentages of CD4(+) T cells with an activated, memory phenotype (effector CD4(+) T cells) in the lamina propria of the intestine compared to peripheral lymphoid tissues, even when examined on the day of birth. Moreover, profound and selective depletion of jejunum lamina propria CD4(+) T cells occurred in neonatal macaques within 21 days of infection, which was preceded by large numbers of SIV-infected cells in this compartment. Furthermore, neonates with less CD4(+) T-cell depletion in tissues tended to have higher viral loads. The persistence of intestinal lamina propria CD4(+) T cells in some neonates with high viral loads suggests that increased turnover and/or resistance to CD4(+) T-cell loss may contribute to the higher viral loads and increased severity of disease in neonatal hosts.  相似文献

Simian immunodeficiency virus (SIV) SIV(smm) naturally infects sooty mangabeys (SMs) and is the source virus of pathogenic infections with human immunodeficiency virus type 2 (HIV-2) and SIV(mac) of humans and macaques, respectively. In previous studies we characterized SIV(smm) diversity in naturally SIV-infected SMs and identified nine different phylogenetic subtypes whose genetic distances are similar to those reported for the different HIV-1 group M subtypes. Here we report that, within the colony of SMs housed at the Yerkes National Primate Research Center, at least four SIV(smm) subtypes cocirculate, with the vast majority of animals infected with SIV(smm) subtype 1, 2, or 3, resulting in the emergence of occasional recombinant forms. While SIV(smm)-infected SMs show a typically nonpathogenic course of infection, we have observed that different SIV(smm) subtypes are in fact associated with specific immunologic features. Notably, while subtypes 1, 2, and 3 are associated with a very benign course of infection and preservation of normal CD4+ T-cell counts, three out of four SMs infected with subtype 5 show a significant depletion of CD4+ T cells. The fact that virus replication in SMs infected with subtype 5 is similar to that in SMs infected with other SIV(smm) subtypes suggests that the subtype 5-associated CD4+ T-cell depletion is unlikely to simply reflect higher levels of virus-mediated direct killing of CD4+ T-cells. Taken together, this systematic analysis of the subtype-specific features of SIV(smm) infection in natural SM hosts identifies subtype-specific differences in the pathogenicity of SIV(smm) infection.  相似文献

Kapusinszky et al. (J Virol 89:8152–8161, 2015, http://dx.doi.org/10.1128/JVI.00671-15) report that host population bottlenecks may result in pathogen extinction, which provides a compelling argument for an alternative approach to vaccination for the control of virus spread. By comparing the prevalence levels of three viral pathogens in two populations of African green monkeys (AGMs) (Chlorocebus sabaeus) from Africa and two Caribbean Islands, they convincingly show that a major host bottleneck resulted in the eradication of select pathogens from a given host.  相似文献
